摘要
以单甲氧基聚乙二醇甲酸琥珀酰亚胺酯(Succinimidyl carbonate methoxy polyethylene glycol,SC-mPEG)为修饰剂对牛血红蛋白(Bovine hemoglobin,BHB)进行了修饰,考察了不同条件下,SC-mPEG与牛血红蛋白反应的产率,并且建立了SC-mPEG-BHB混合物的分析方法。根据不同反应因素对SC-mPEG修饰血红蛋白反应的影响确定了SC-mPEG与牛血红蛋白反应的最佳条件,即SC-mPEG与牛血红蛋白最佳配比为60∶1,在4℃,pH9的缓冲溶液体系反应2h。
This paper aims to raise concerns about the modification of bovine hemoglobin with the use of SCmPEG as modifying reagent. The yield rates of SC-mPEG-BHB under dissimilar conditions are studied, and more important, a series of methods used to characterize the SC-mPEG--BHB mixtures are built. The best reactive system has been confirmed, i.e. at 4℃, pH9; SC-mPEG and BHB are mixed according to the proportion of 60: 1 and be submitted to reaction for 2 h.
