摘要
目的探讨牛磺酸对液压冲击伤模型神经细胞伤后不同时间细胞内游离钙超载的保护作用。方法采用激光扫描共聚焦显微镜观察液压冲击伤模型伤后24 h和48 h神经细胞形态变化,并检测细胞内游离钙表达水平变化。结果损伤后,神经细胞肿胀,体积增大,突起回缩,细胞内游离钙表达水平升高,于伤后24 h达峰值水平,至伤后48 h仍维持在较高水平且高于对照组(均P<0.01)。经牛磺酸治疗后,神经细胞肿胀明显改善,接近正常细胞大小,突起回缩程度减轻,细胞内游离钙表达水平逐渐降低(均P<0.01),且各时间点均低于创伤组(P<0.01),以伤后1 h牛磺酸治疗48 h组效果最佳,细胞内游离钙表达水平接近正常值水平(P>0.05)。结论牛磺酸对液压冲击伤模型神经细胞内钙超载具有较好的保护作用,尤以伤后1 h予以牛磺酸并维持治疗48 h对神经细胞保护作用最佳。
Objective To study the protective effect of taurine on calcium overload in rat neurons after hydraulic impact injury at different times. Methods The cultured rat neurons were randomly divided into 3 groups: normal control group (control group), hydraulic impact injury group (injury group) and taurine treatment group (treated with taurine immediately or 1 h after injury, taurine group). Laser scanning confocal microscope (LSCM) was used to examine neuronal histomorphological changes 24 h and 48 h after injury, and expression level of intracellular [Ca^2+]i was measured with Fluo-3/AM as a calcium indicator. Results After injury, the nerve cells became swelling and larger, and the process retracted. The expression level of intracellular [Ca^2+]i increased, achieved to peak at 24 h, and still remained on high level at 48 h after injury. The expression level of intracellular [Ca^2+]i in injury group was higher than that in controll group (P〈 0.01, for all). After taurine treatment, the neuronal swelling was improved obviously, and the neuron size almost approached to normal, while the process retraction reduced. The expression level of intracellular [Ca^2+]i in taurine group gradually decreased (P〈0.01, for all), and was lower than that in injury group at every time point (P〈0.01, for all). The optimum effect was seen in 48 h-traurine group when treatment was given 1 h after injury, and the expression level of intracellular [Ca^2+]i approached to normal (P〉 0.05). Conclusion Taurine is effective in counteracting the calcium overload in rat neurons caused by hydraulic impact injury. It is most effective when the treatment is given 1 h after injury and sustained for 48 h.
