摘要
目的:建立芒果苷的高效液相色谱测定法,研究其在Caco-2细胞上的转运特征。方法:利用MTT实验筛选出芒果苷在Caco-2细胞中的安全浓度,然后用Caco-2细胞单层模型研究芒果苷的转运特征。采用HPLC法检测药物浓度,考察时间、药物浓度、P-糖蛋白抑制剂对芒果苷在Caco-2细胞上转运的影响。结果:芒果苷在Caco-2细胞单层膜上的转运量具有一定的浓度和时间依赖性,P-糖蛋白抑制剂维拉帕米和环孢素A能够增加芒果苷跨膜转运量。结论:芒果苷在Caco-2细胞上转运表现为被动扩散且有外转运蛋白参与芒果苷跨膜转运。
OBJECFIVE To establish an HPLC method for determination of mangiferin and investigate its transport in Caco- 2 cell monolayer model. METHODS The safety concentration of mangiferin in Caco-2 cell was first selected by using MTT method. Then the Caco-2 cell monolayer model was used tO investigate the transport of mangiferiru An HPLC method was applied to determine mangiferin concentration in Caco-2 cell monolayer transport system under various conditions, including changing mangiferin concentration and time. Furthermore, influence of positive P-gp inhibitor veraparnil on mangiferin transport was involved in the study. RESULTS Mangiferin transport in Caco-2 cell monolayer was time and concentration dependent. The transport of mangiferin was enhanced by verapamil and cyclosporine A. CONCLUSION Mangiferin transport complied with the passive diffusion in Caco-2 cell monolayer model, and furthermore efflux transporters might be involved in the transepithelial transport of mangiferin.
