摘要
目的:探讨肝癌中Survivin、PTEN和NF-κB蛋白的表达与临床病理参数的关系。方法:用EliVisionTM plus免疫组化方法检测Survivin、PTEN和NF-κBp65蛋白在30例肝癌、癌旁组织及10例正常肝组织中的表达情况。结果:Survivin及NF-κBp65蛋白在肝癌中表达分别为60.00%、66.67%;显著高于癌旁组织(23.33%、13.33%)及正常肝组织(0、10.00%)(均P<0.0125);PTEN蛋白在肝癌、癌旁及正常肝中表达分别为56.67%、93.33%、100%,其中肝癌组织的表达水平显著低于癌旁组织和正常肝组织(均P<0.0125);但癌旁组织与正常肝组织比较差异无统计学意义(P>0.05)。Survivin、PTEN蛋白的表达与门脉浸润密切相关(P<0.05),肝癌组织中Survivin蛋白表达与PTEN蛋白表达呈负相关,与NF-κBp65蛋白表达呈正相关(P<0.05)。结论:Survivin、PTEN和NF-κBp65蛋白异常表达与肝癌的发生和发展有密切关系。Survivin激活、上调和PTEN失活、下调可能参与肝癌的发生发展;Survivin与NF-κBp65蛋白可能在肝癌的发生发展中起协同作用。
Objective:To investigate the protein expression of Survivin,PTEN and NF-κB genes in hepatocellular carcinoma(HCC) tissues and the correlation between their clinic pathologic parameters.Methods:Immunohistochemical technique was used to detect the protein expression ratio and intensity of Survivin,PTEN and NF-κBp65 in 30 specimens of HCC,surrounding nonmalignant hepatic tissues and 10 specimens of normal liver tissues.Result:The expression of Survivin protein and NF-κBp65 protein in human HCC was 60.00%,66.67%.Both of them were significantly higher than surrounding nonmalignant hepatic tissues(23.33%,13.33%) and normal liver tissues(none,10.00%)(both P〈0.012 5).The expression of PTEN protein in human HCC,surrounding nonmalignant hepatic tissues and normal liver tissues was 56.67% (17/30),93.33%(28/30) and 100%(10/10),respectively; The expression of PTEN protein in human HCC was markedly lower than surrounding nonmalignant hepatic tissues and normal liver tissues(all P〈0.012 5); But it was not statistically significant difference between surrounding nonmalignant hepatic tissues and normal liver tissues (P〉0.05).The expression of Survivin and PTEN protein in HCC was significantly correlated with portal vein invasion(P〈0.05). Survivin protein expression in HCC was inversely associated with PTEN protein expression (P〈0.05) and was positive correlation with NF-κBp65 protein (P〈0.05).Conclusion:The abnormal expression of Survivin,PTEN and NF-κBp65 protein in HCC sugvip that they probably play important roles in the origin and development of HCC. Survivin activation,up-regulation and PTEN inactivation,down-regulation probably participate in the origin and development of HCC. Survivin protein and NF-κBp65 protein may have a synergistic effect in the origin and development of HCC.
