实时定量反转录-聚合酶链反应检测机械力对人牙周膜细胞骨钙素表达的影响

摘要 目的观察周期性牵张力作用下人牙周膜细胞(HPDLCs)成骨分化关键基因骨钙素的表达变化,以明确周期性牵张力对HPDLCs成骨分化的诱导作用。方法利用自行研制的多通道细胞牵张应力加载系统对体外培养的HPDLCs施加12%形变率、0.1Hz周期性牵张力1,2,3,5d,实时定量反转录-聚合酶链反应(RT-PCR)检测其成骨分化基因骨钙素(BGLAP)表达的时序性变化。结果体外培养HPDLCs骨钙素基因表达量极低,加力1、2、3、5d后骨钙素基因表达依次升高,并且在加力5d时表达量明显增高为对照组的71.6倍。结论周期性牵张力可以诱导HPDLCs向成骨方向分化,骨钙素在机械力诱导成骨分化的后期发挥作用。 Objective To investigate the effects of cyclic tension force on the expression of BGLAP in human periodontal ligament cells. Method Human periodontal ligament cells (HPDLCs) were subjected to cyclic tension force for 1,2,3 and 5 days and the control cells without application of tension force. The mRNA expressions of later differentiation gene (osteocalcin/BGLAP) were detected by real-time RT-PCR. Results HPDLCs expressed BGLAP gene very lowly in vitro. The expression of BGLAP gene was up regulated after application of tension force for 1, 2, 3 and 5 days. The expression of BGLAP gene at day 5 was 71.6 times to the control cells. Conclusion Cyclic tension force can induce the osteogenic differentiation of human periodontal ligament cells and increase the expression of BGLAP in the later differ-entiation stage.
