摘要
以玉米幼叶生长部位为材料,通过水溶性多聚物PEG4000/DextranT500构成的两相系统,对玉米幼叶生长部位质膜进行了分离。结果表明,由PEG40006.2%、DextranT5006.2%、KCl5mmol/L、磷酸缓冲液pH7.84mmol/L、蔗糖8.5%构成的两相系统最适宜分离玉米幼叶生长部位质膜。经过三次相分配后,质膜在上相中的分配率达85%以上。经-0.4MPa胁迫处理24h后的材料在上述两相系统中分配时,其质膜、叶绿体膜、线粒体膜在上相中的分配率均稍有下降,内质网膜、液泡膜则稍有增加,但这些变化都不太显著,并且质膜在上相中的分布占绝对优势。
The Plasma Membranes (PM) of maize young leaves growth zone were isolated by the method of two phase partition.The results showed that the best composition of two phase system which isolated PM was:Dextran T500 6.2%,PEG 4000 6.2%,K phosphate buffer pH 7.8 4 mmol/L,KCl 5 mmol/L and sucrose 8.5%.After three times phase partition,the partition of PM in the upper phase was beyond 85%.When maize seedlings were stressed by -0.4 MPa for 24 hour,the partition of PM and other internal membranes in the upper phase took some changes,but the changes were not obvious,and PM was in advantage of the upper phase absolutely.
