摘要
目的:研究天花粉蛋白诱导小鼠前列腺癌细胞RM-1凋亡的作用,并探讨其作用机制。方法:将不同浓度的天花粉蛋白作用于小鼠RM-1前列腺癌细胞,用MTT法检测其半数抑制浓度(IC50),用这一浓度作用于RM-1细胞一定时间后,通过Hoechest33258荧光染色、流式细胞仪检测RM-1细胞凋亡的情况。采用Western Blot检测凋亡基因bax蛋白表达的变化。结果:MTT法测得天花粉蛋白对RM-1细胞的IC50值为117.32 mg/L;Hoechest33258荧光染色提示天花粉蛋白干预后RM-1细胞出现凋亡小体;流式细胞仪检测结果显示随着天花粉蛋白干预时间的延长,RM-1细胞凋亡率增加,同时凋亡基因bax蛋白的表达在天花粉蛋白干预后逐渐增加。结论:天花粉蛋白具有抗小鼠前列腺癌细胞RM-1的作用,其作用机制之一是诱导肿瘤细胞凋亡;bax基因在天花粉蛋白诱导RM-1细胞凋亡过程中起重要调控作用。
Objective:To study the inducing effect of trichosanthin on the apoptosis of mouse prostatic cancer RM-1 cells and its mechanism. Methods : MTT assay was used to identify the effect of trichosanthin on RM-1 cells in vitro. The cells apoptosis was detected by Hoeehest 33258 fluorescent staining and flow cytometry. The levels of bax proteins were detected by western blot analysis as well as their apoptosis rate. Results:The IC50 value of trichosanthin for RM-1 cell was 117.32 mg/L Cell cycle was analysised by flow cytometry, trichosanthin induced an arrest in G0/G1 phase. Hoechest 33258 fluorescent staining showed typical nucleolus changes in apoptosis cells. Expression of bax was up-regulated at protein levels in a time-dependent manner. Conclusion:Triehosanthin can induce apoptosis in RM-1 cells. The induction of apoptosis is a very important mechanism of trichosanthin to inhibit prostatic cancer.
