摘要
目的研究五倍子水提取物对金黄色葡萄球菌生物膜的影响。方法采用试管二倍稀释法测定受试药物的最低抑菌浓度(minimal inhibitory concentration,MIC),用平板改良法体外模拟金黄色葡萄球菌生物膜模型,并检测五倍子水提取物不同的MIC浓度对金黄色葡萄球菌生物膜的影响作用,扫描电镜确认。结果五倍子水提取物对金黄色葡萄球菌的MIC值为64 mg/mL,当增加药物浓度至1.5倍MIC(96 mg/mL)和2倍MIC(128 mg/mL)分别于4、8和24 h作用金黄色葡萄球菌生物膜时,活菌数明显减少,与生理盐水对照组比较,差异有显著性(P<0.05),但24h内金黄色葡萄球菌BF中的细菌不能被完全清除。各时间段之间活菌数差异没有显著性(P>0.05)。结论五倍子水提取物对金黄色葡萄球菌生物膜有清除影响,具备开发应用的潜力,但还需进行药物配方和临床用药研究。
[Objective] To study resistance of Staphylococcus aureus Biofilm to Gallnut water extract compound preparation. [Methods] Minimal inhibitory concentration (MIC) was determined by use of method of doubling dilution, while simulate S. aureus biofilm in vitro was analyzed by improved Brown plate method. The effects of Gallnut water extract in different MIC on S. aureus biofilm were determined and confirmed by scanning electronic microscope (SEM). [Results] The MIC of extracted Gallnut water to S. aureus was 64 mg/mL, when the MIC reached 1.5 ×MIC (96 mg/mL) or 2 × MIC (128 mg/mL) acted on S. aureus biofilm for 4 hours, 8 hours and 24 hours respectively, the count of active S. aureus bio-film was significant difference (P 〈0.05) compared with normal saline control group. However, the bacteria in S. aureus Biofilm were not completely eliminated within 24 hours, and there was no significant difference (P 〉0.05) in viable count of S. aureus bio-film in the 3 different time. [Conclusion] Gallnut water extract compound preparation had ability to eliminate the S. aureus biofilm, thus potential to be exploited. But it also needed to be further studied in formulation and clinic application.
