摘要
目的:研究结缔组织生长因子的mRNA在细菌感染导致的慢性阻塞性肺疾病大鼠肺组织中的表达。方法:以适量肺炎链球菌反复经气管穿刺注射感染大鼠,制作大鼠慢性阻塞性肺疾病模型,在感染后的不同时间段处死大鼠,采用实时荧光定量PCR,对大鼠肺组织中结缔组织生长因子的mRNA表达情况进行研究。结果:生理盐水对照组结缔组织生长因子mRNA的表达明显低于肺炎链球菌组(P<0.01)。同时,肺炎链球菌组结缔组织生长因子mRNA的表达随着细菌感染时间的增加而升高(P<0.01)。结论:大鼠长期反复细菌感染可诱导慢性阻塞性肺疾病,其机制可能和结缔组织生长因子表达上调有关。
Objective To explore the expression of mRNA of connective tissue growth factorc (CTGF) in the pulmonary tissues of rats with chronic obstructive pulmonary disease (COPD) induced by bacterial infection. Methods The rat model of COPD was established by repeated intratracheal injections of streptococcus pneumoniae. The expression of mRNA of CTGF in the pulmonary tissues was detected by real-time PCR at different time points after establishment. Results The expression of mRNA of CTGF was obviously lower in the control group than in the model group (P 〈 0.01 ) ; meanwhile, the mRNA expression in the latter group was increased in a time-dependent manner (P 〈 0.01). Conclusion Repeated bacterial infections can result in COPD in rats, whose mechanism may be associated with the upregulation of CTGF expression.
