摘要
采用单因子实验和正交试验对具抑菌活性放线菌JSM20.1发酵培养基和发酵条件进行优化,并对其产生的抑菌活性物质的特性进行了初步研究。优化后的最佳发酵培养基:葡萄糖15.0 g,大豆粉10.0 g,NaCl 5.0 g,酵母膏1.0 g,CaCO31.0 g,复合盐A液20 mL,复合盐B液1 mL,水1000 mL,最佳起始pH7.0。优化后的最佳发酵条件:摇瓶装量20%,摇床转速200 r/min,发酵温度28℃。所产生的抑菌活性物质有较好的热稳定性,在强酸、强碱和高温条件下都具有稳定活性,不溶于石油醚和氯仿,溶于乙酸乙酯。
Single-factor experiment and orthogonal tests were adopted to optimize the fermentation medium and conditions for Streptomyces sp. JSM20. 1 that exhibited strong bacteriostasis activity, the characters of its antibiotic substance were studied as well. After the optimization, the optimum fermentation medium was glucose 15.0 g, soybean flour 10 g, NaCl 5.0 g, yeast extract 1.0 g, CaCO3 , 1.0 g, broth A of compound salts 20 mL, and broth B of compound salts 1 ml, water 1 000 mL, optimal starting pH at 7.0. After the optimization, the optimal shaking flask filled by 20% , rotated at 200 r/min, fermentation temperature 28 ℃. The product had fairly stable against heat, and showed stable activity under strong acid, alkali, and high temperature, insoluble in petroleum ether and chloroform, soluble in ethyl acetate.
