摘要
目的:了解阿司咪唑对心脏收缩功能的影响。方法:运用Langendorff灌流装置逆行灌流大鼠体外心脏,测定阿司咪唑对左心室内压的影响;运用器官浴槽系统观察阿司咪唑对乳头肌和心房肌肌张力的影响。结果:随着剂量增加和作用时间延长,阿司咪唑降低左心室平均收缩压和最大收缩压的作用增强,明显减慢左心室内压等容相上升最大速率(+dp/dtmax);阿司咪唑还能明显降低乳头肌和心房肌的收缩张力,使乳头肌收缩张力+dp/dtmax随药物浓度增加而逐渐减慢。结论:阿司咪唑能明显降低心肌收缩力,对大鼠体外心脏的收缩功能有显著抑制作用。
Objective:To explore the effect of Astemizole on systolic function of heart in rats. Method:Langendorff device was used to perfuse inversely isolated heart of rats and the effects of Astemizole on intraventricular pressure was determined. The muscular tension of free papillary muscle and auricle muscle were recorded by RM6240BD-type gather equipment of physiology signal. Result: Astemizole depressed the mLVSP, LVSPmax and +dP/dtmax of left ventricle and the action was enhanced along with the increasing of astemizole dose and the lasting of action time. Astemizole also weakened the contraction force of free papillary and auricle muscle obvioushy, slowed +dT/dt max of free papillary muscle gradually with the increasing dose. Conclusion: Astemizole can weakened obviously myocardial contractility, and inhibit observably the contraction function of rat isolated heart.
