摘要
目的:检测大肠癌组织中神经纤毛蛋白-1(Neuropilin-1,NRP-1)mRNA的表达情况及微血管密度(MVD)值,探讨NRP-1mRNA的表达与MVD间的相关性,以期了解NRP-1与肿瘤血管生成的关系。方法:取45例大肠癌组织及相应正常大肠组织,采用半定量RT-PCR方法检测其NRP-1mRNA的表达情况,采用免疫组化染色法检测CD105标记的新生肿瘤血管,计数MVD值。结果:45例大肠癌组织中39例可见NRP-1mRNA的表达,阳性率为87.0%;相应正常大肠组织仅3例表达,阳性率为6.7%。大肠癌组织中NRP-1mRNA的表达程度与肿瘤的大小、浸润的深度、淋巴结转移呈正相关(P<0.05),与肿瘤部位、组织学类型、分化程度、患者的性别及年龄无关(P>0.05)。大肠癌组织中NRP-1mRNA的表达程度随MVD值的增加而升高(r=0.38,P=0.01)。结论:NRP-1可能与大肠癌的肿瘤血管生成相关,NRP-1mRNA高表达代表着大肠癌不良的生物学行为趋向。
To study the expression of NRP-1mRNA and detect microvessel density (MVD) in colorectal cancer, and investigate the correlation between NRP-1 mRNA and MVD to know the role of NRP-1 in the process of tumor angiogenesis. Methods :In this study, 45 specimens of colorectal cancer tissue and normal tissue were used simultaneously. The expression of NRP-1 mRNA in colorectal cancer was determined by means of reverse transcription-polymerase chain reaction (RT-PCR) and MVD was figured out based on CD105 immunohistochemical staining. Results:In this study, over expression of NRP-1mRNA was found in colorectal cancer. The positive case of NRP-1mRNA was 87. 0% (39/45) in colorectal cancer, but in normal colon tissue was 6. 7% (3/45). The expression of NRP-1 mRNA had a good correlation with the tumor size, infiltrating depth, lymph node metastasis (P 〈 0. 05 ), but no correlation was found with the tumor position, histological types, differentiation degree and the gender or age of patients (P 〉 0. 05 ). A close positive correlation was present between NRP-1 mRNA and MVD (r = 0. 38,P = 0. 01 ). Conclusion :NRP-1 may play a part in the angiogenesis of the colorectal cancer and the high expression of NRP-1mRNA represented the malignant biological behavior trend.
