摘要
目的:建立紫外分光光度法测定血液中盐酸罂粟碱含量的方法。方法:血液样品中加入KOH溶液调节pH至12,振荡后加入三氯甲烷-正己烷(2:3)作为萃取剂,振荡30 min,取上层部分提取液于60℃水浴蒸干,残渣溶于1 ml1 mol/L HCl溶液中,用0.45μm滤膜过滤后在波长250.4 nm下紫外分光光度计测定提取液的吸光度,建立了一种简单、适用的血液盐酸罂粟碱检测方法。结果:方法检出限为0.05 mg/L,标准曲线线性相关系数0.9989,样品加标回收率在85.7%~91.3%之间,日内及日间RSD均在3.2%~8.6%之间。测定了兔子静脉注射盐酸罂粟碱注射液后不同时间内血液中盐酸罂粟碱的含量,对其药时曲线进行了研究。结论:该方法准确、稳定,有良好的重现性和较高的灵敏度。
Objective:A simple and adequate analysis method is described for the determination of papaverine hydrochloride in blood using the conventional spectrophotometry.Methods:The pH of blood samples were adjusted to about 12,and chloroform-n-hexane(2:3) as the extracted solvent was used.After centrifugation,partial organic phase was retrieved and dried in 60℃ water bath.The dried product was dissolved in 1 ml 1.0 mol/L HCl and then filtrated through 0.45 μm membrane.The filter extracts was detected directly by UV-vis spectrophotometry at 250.4 nm wavelength.Results:The detection limit was 0.05 mg/L.The correlation coefficient for the standard curve was 0.9989,and the recoveries of papaverine hydrochloride 85.7%~91.3%.The RSD values of within-day and between-day precision were 3.2%~8.6%.The method was used in analysis of papaverine hydrochloride concentration in rabbit vein blood after injecting papaverine hydrochloride injection.Conclusion:The proposed method is precise and suitable for the detection of papaverine hydrochloride in blood serum and has been successfully applied in a concentration via time curve study.
