摘要
从湖南岳阳南湖、同联药业和城陵矶等3个洞庭湖样点采集表层水样,分别编号为NH、TL和CL.在进行水样元素成分分析的同时,采用扩增rDNA限制性酶切片段分析(Amplifed Ribosomal DNA Restriction Analysis,ARDRA),对上述水样中的细菌多样性进行研究.3个样品共挑出234个克隆子进行酶切,得到135个可操作分类单元(Operational Taxonomic Units,OTUs),并对其中14个优势克隆子进行测序.系统发育分析显示洞庭湖岳阳段水样中微生物群落具有丰富的多样性,其中:NH样品中的优势菌是Flavobacteriumsp和Pseudomonas sp,分别占22.4%和11.2%;CL样品中的优势菌为Catellibacterium和Acinetobacter,分别占9.6%和8.2%;而样品TL中的优势菌则为属于β-Proteobacteria的一个新菌株,占11.1%.
Three water samples were collected from Nanhu (NH), Tonglian pharmaceutiacal company(TL) and Chenglingji (CL) in Yueyang, Hunan. In addition to element analysis, the bacterial diversity was described by amplified ribosomal DNA restriction analysis (ARDRA). A total of 234 clones were obtained, 135 ARDRA patterns were found and 14 dominant clones were selected for sequencing. Phylogenetic analysis showed that there was great bacterial diversity among the three water samples. The dominant bacteria of sample NH were Flavobacterium sp and Pseudomonas sp, represented 22.4% and 6.8% respectively. The dominant bacteria of sample CL were Catellibacterium and Acinetobacter, represented 9.6% and 8.2% respectively. The dominant bacterium of sample TL was a new strain belonging to β- Proteobacteria, represented 11.1%.
