摘要
本实验利用激光这一高效诱变手段,对野生菌株绿色木霉Tr-02进行辐照诱变,通过两轮的定向筛选,最终得到一株高产木聚糖酶的菌株。对突变株进行发酵条件优化,结果表明,以0.5%麸皮为碳源,0.5%蛋白胨为氮源,0.1% Tween-80作为产酶促进剂,培养基初始pH值为5.0,瓶装量100mL,接种两环,28℃,180r/min培养48h,木聚糖酶活力达到281.21U·mL-1,较之相同条件出发菌株木聚糖酶活力增长31.83%。
A mutation of wild strain Trichoderma Viride Tr-02 irradiated by laser was studied in this paper. A high xylanase-producing strain was obtained by two steps oriental screening finally. The results show that an optimization condition of the xylanase fermentation is as following: 0.5% corncob as the carbon source, 0.5% peptone as the nitrogen source, 0.1% Tween-80 as a enzyme-promoting agent, initial medium pH to 5.0, medium volume to 100mL, vaccinated two rings of spores, 28℃, 180 r/min, fermented 48h. The xylanase activity of the mutant reaches 281.21U/mL and is 31.83% higher than that of the original strain under the same conditions.
