摘要
宫颈癌是女性最常见的一种恶性肿瘤,研究表明人乳头瘤病毒(HPV)感染与宫颈癌的发生密切相关.采用实时荧光PCR技术对引起宫颈癌的两种主要HPV亚型HPV 16和18型阳性质粒进行检测,在实验室中成功构建了HPV 16和18型的检测体系以及体外模拟的双重感染检测体系,并对三例临床确诊为宫颈癌的标本进行实时荧光PCR验证检测,确定三例标本均为高浓度HPV 16型感染,极易引起宫颈癌变,与医院诊断结果相符合.本实验为实时荧光PCR技术检测HPV 16和18型的临床检验在实验室中成功地进行了探索,为实时荧光PCR技术在宫颈癌大规模筛查上的应用提供了研究基础.
Cervical carcinoma is one of the most common cancers in women, most of cervical carcinoma is caused by HPV (Human Papillomavirus). HPV16 and HPV18 are two main high risk subtypes causing cervical carcinoma. HPV16 and HPV18 DNA fragments are detected by the realtime PCR with the successful sub-typing detection system designed with positive plasmids. Another dual-infection detection system is designed for sub-typing in dual-infection. The system is confirmed by three tissue samples from operations. The method of detection HPV by real-time PCR is proved to be effective in practice, a rapid, sensitive and valuable assay to detect HPV16 and HPV18 infections with cervical cells ,can be applied for large-scale screening and clinical diagnosis.
