摘要
目的:探讨NK、T混合淋巴细胞(NKTm)与顺铂联合应用于肺巨细胞癌PLA-801D细胞的最佳时序方案,为临床治疗提供实验依据。方法:按加入NK、T混合淋巴细胞(NKTm)和顺铂的时序不同分为7个时序组,每个时序组中设单顺铂组,单NK、T混合淋巴细胞(NKTm)组及联合组,MTT法分别测定各组对PLA-801D细胞的抑制率。结果:各时序组中联合组的抑制率高于单顺铂组和单NK、T混合淋巴细胞(NKTm)组(P<0.01),各时序组联合应用的Q值均大于1.15,且先用NK、T混合淋巴细胞(NKTm)4 h后再用顺铂组的Q值最大(Q=2.297)。结论:体外研究表明,NK、T混合淋巴细胞(NKTm)与顺铂联合应用具有协同效应,先用NK、T混合淋巴细胞(NK-Tm)后4 h再用顺铂为最佳时序方案。
Objective: To investigate the best choice of combined treatment to human pulmoal giant cell carcinoma cells (PLA-801 D)with mixture of NK and T lymphocytes (NKTm cells)and cisplatin (DDP) in different sequences. Methods: The inhibited rates of PLA-801D cells treated by single NKTm cells, single DDP, and combined treatment of NKTm cells and DDP in different sequences were determined with MTr colorimetric assay. Results: The effect of combined treatment of NKTm ceils and DDP excelled the effect of single NKTm cells treatment and single DDP treatment(P 〈 0.01 ). Values of Q in all the combined group were more than 1.15 ; among all the group, the sequence of using NKTm ceils first, and then 4 h later using DDP had the highest Q value( Q = 2.297). Conclusion: Combined treatment of NKTm cells and DDP had synergistic effect to inhibit PLA- 801D cells. The best sequence was to using NKTm cells first, and then 4 h later using DDP.
