葡萄试管苗不定根发生发育的组织学和细胞学研究 被引量:7

A study on Histology and Cytology of Origin and Development of Adventitious Root of Grape Plantlets in Testtuble

摘要 通过对葡萄试管苗不定根发生发育过程的外部形态及内部组织解剖研究发现,藤稔与全球红两品种葡萄在离体茎段培养诱导生根中表现差异较大,全球红在外源激素的诱导下能很好地发出不定根,而藤稔则诱导不定根较为困难,个体生根数目差异较大,且诱导出的不定根生长缓慢,愈伤组织大量生长。利用组织切片观察它们在根原基发生发育各个时期的内部组织结构,发现两品种不定根根原基起源部位相同,都是在形成层与髓射线交接处形成不定根原基,培养第6~8天开始不定根原基的分化,此后逐步进行根的组织分化。不定根的维管组织与茎段中的维管组织衔接很好。外观上的差异只是因为不同品种对激素的敏感程度不同。 The histological and morphogical study of origin and development of adventitious root of grape plantlet in testtube was conducted.It showed that the two varieties “Fujiminori” and “Whole Globe Red” were significant difference in adventitious root inducing of in vitrostem culture.The adventitious root could be generated very well with exogenous hormone on “Whole Globe Red”,but “Fujiminori” was comparatively difficult to induce adventitious root,the number of individual roots was more different.The induced root grew slowly and the callus grew in large quantity.The histological section observation in each period of origin and development of adventitious root indicated that the location of adventitious root origin was the same in the two varieties.It was connecting place of cambium and pith ray that the adventitious root primordium was formed.The adventitious root primordium differentiated on the 6th to 8th days.Later the tissue differentiation of the roots gradually appeared.The vascular tissue of the root and the stem joined quite well.The differences of exterior were just due to the different sensibility between two varieties.
