铁离子增强青蒿琥酯抗癌作用及改变癌细胞死亡方式研究 被引量:4

摘要 目的研究青蒿琥酯的抗肺腺癌细胞的作用与全铁转运蛋白的关系以及青蒿琥酯诱导细胞死亡的方式。方法利用MTT比色法观察不同质量浓度青蒿琥酯对培养的肺腺癌细胞在应用或不用全铁转运蛋白预处理的情况下48 h后的抑制率和IC50;并通过细胞形态学观察、DNA电泳、流式细胞仪分析肺腺癌细胞的死亡方式。结果48 h单纯青蒿琥酯组对肺腺癌的IC50为53.21μg/mL,用全铁转运蛋白处理后的IC50为12.41μg/mL;单纯青蒿琥酯组,48 h后细胞体积缩小,新月体形成,DNA电泳呈梯形,流式细胞仪分析显示亚G1峰;用全铁转运蛋白处理后,24 h细胞体积增大肿胀,染色质分散、边集,DNA电泳仅在1 000 bp和2 000 bp见条带,流式细胞仪分析无亚G1峰,与对照类似。结论全铁转运蛋白可增强青蒿琥酯的抗肺腺癌细胞的作用,加快癌细胞的死亡速度,两者联合可诱导癌细胞以胀亡的方式死亡。
