Expression of SOX9 in colorectal cancer and its significance

摘要 目的探讨SOX9、SOX8在结直肠肿瘤中的表达情况及其预后意义。方法应用免疫组化EnVision法检测226例结直肠癌及41例腺瘤中SOX9、SOX8的表达,并分析蛋白表达与临床病理参数间的关系。结果SOX9、SOX8在结直肠肿瘤中均有表达。在正常黏膜、癌旁黏膜、腺瘤和腺癌中,SOX9的表达率逐渐增高,分别为9.1%(10/110)、14.0%(16/114)、48.8%(20/41)和60.1%(113/188)(P<0.01),SOX8阳性表达率为0%(0/117)、0%(0/125)、29.3%(12/41)和15.2%(31/204)。SOX9高表达仅见于结直肠肿瘤。SOX9在黏液腺癌和印戒细胞癌等中表达相对较低,而SOX8表达则主要见于黏液腺癌和印戒细胞癌。Kaplan-Meier生存曲线分析证实结直肠癌SOX9高表达为预后不良,COX比例风险模型证实SOX9高表达是结直肠癌的独立预后因素(P<0.05)。结论SOX9在结直肠癌高表达,且高表达是结直肠癌预后不良指标。 Objective To investigate the expressions of SOX9 and SOX8 in colorectal cancer and its correlation with the clinopathologic parameters. Methods The expressions of SOX9 and SOX8 were detected with immunohistochemical staining in 226 cases of colorectal cancer and 41 of sporadic adenoma. Results Nuclear staining of SOX8 and SOX9 was found in colorectal tissues. The incidence of SOX9 expression increased sequentially from normal mucosa (9.1%, 10/110), peri-cancer mucosa (14.0%,16/114) and adenoma (48. 8%,20/41) to cancers (60. 1%, 113/188)(P〈0. 01). SOX8 expression was absent in normal mucosa and peri-cancer mucosa, but SOX8 was expressed in adenomas (29. 3%, 12/41) and cancers (15.2%, 31/204). SOX9 overexpression (≥++) was only found in colorectal adenomas and cancers (P〈0. 01). SOX9 overexpression in mucinous/signet-ring cell carcinoma was uncommon while SOX8 expression was primarily in mucinous/ signet-ring cell carcinoma (P〈0. 01). Both SOX8 and SOX9 expressions were not associated with other clinopathologic parameters including histology grading (P〉0.05). It was also found that SOX9 overexpression was an independent indicator of adverse prognosis for colorectal cancer (P〈0. 01). Conclusions SOX9 expression is upregulated in colorectal cancers and its overexpression is an independent adverse prognosticator in colorectal cancers.
