摘要
目的通过建立大鼠急性高眼压模型,检测孕酮(PROG)对急性高眼压下大鼠视网膜神经节细胞的保护作用。方法选取健康Wistar大鼠35只,分3组,Ⅰ组即正常组5只(10眼),Ⅱ组即孕酮腹腔注射组15只和Ⅲ组即生理盐水腹腔注射组15只,Ⅱ、Ⅲ组左眼采用前房加压灌注法制成急性高眼压模型,右眼为自身对照眼,最终弃去不符合标准的眼球,每组取10只眼球固定后行石蜡切片,做HE染色及Thy-1.1免疫组化、TUNEL原位凋亡检测。结果Ⅱ、Ⅲ组模型眼的视网膜神经节细胞数均较I组和自身对照眼明显减少(P<0.05);但Ⅱ组模型眼视网膜神经节细胞数多于Ⅲ组模型眼(P<0.05);Thy-1.1阳性细胞数Ⅱ、Ⅲ组模型眼均低于对照眼(P<0.05);但Ⅱ组模型眼Thy-1.1阳性细胞数多于Ⅲ组模型眼(P<0.05)。TUNEL染色阳性细胞数Ⅱ、Ⅲ组模型眼均高于对照眼(P<0.05),Ⅱ组模型眼TUNEL染色阳性细胞数少于Ⅲ组模型眼(P<0.05)。结论孕酮对大鼠急性高眼压模型的视网膜神经节细胞有保护作用。
Objective To observe the neuroprotection of PROG on rat retinal ganglion cells in acute ocular hypertention model. Methods 35 Wistar rats without eye diseases were divided into 3 groups: group Ⅰ :normal group(5rats, 10eyes), group Ⅱ :PROG treated group (15 rats),groupⅢ :balanced salt solution treated group( 15 rats).Rats in group Ⅱ and Ⅲ were induced in left eye by forcing perfusion of balanced salt solution into the anterior chamber as acute ocular hypertenfion model. Right eyes were self-control eyes.Not standard eyeballs were discard and10 eyeballs were selected and fixed in each group.The retinal ganglion cells were detected by HE staining and Thy1 immunohistological technlque.The apoptosis cells were detected by TdT-dUDP terminal nick end-labeling (TUNEL) technique.Results The RGCs quantity of model eyes in group Ⅱ and Ⅲ were less than that of group Ⅰ and self-control eyes, there were significant diferences ( P 〈 0.05 ). But the RC, Cs quantity of model eyes in group Ⅱ were more than that of group Ⅲ, there were significant diferences (P 〈 0.05). The Thy-1.1-positive cells of model eyes in group Ⅱ and Ⅲ were less than that of group Ⅰ and self-centrol eyes,there were significant diferences ( P 〈 0.05). But the Thy-1.1-positive cells of model eyes in group Ⅱ were more than that of group Ⅲ, there were significant diferences ( P 〈 0.05). The TUNEL-positive cells of model eyes in group Ⅱ and Ⅲ were more than that of group Ⅰ and self-control eyes,there were signifieant diferences ( P 〈0.05).But the TUNEL-posifive cells of model eyes in group Ⅱ were less than that of group Ⅲ, there were significant diferences ( P 〈 0.05 ). Condusion PROG could provide neuroprotecfion for RC, Cs in mt acute glaucoma model.
