双歧双歧杆菌细胞壁完整肽聚糖的分离纯化 被引量:38

ISOLATION AND PURIFICATION OF WHOLE PEPTIDOGLYCAN FROM BIFIDOBACTERIUM BIFIDUM

摘要 本试验采用国内分离的双歧双歧杆菌菌株Bif1101,用TritonX-100处理BL肉汤培养物,高速离心分离不可溶沉渣,经蛋白酶E、胃蛋白酶、α-糜蛋白酶、胰酶、核酸酶消化,甲醇与氯仿除脂,0.01NH2SO4处理等步骤提纯而得到精制完整肽聚糖(wholepeptidoglycan,WPG)。化学组分分析结果显示WPG主要由多肽与糖组成,其氨基酸组成与菌体基本相同。扫描电镜观察发现提取的WPG保留其原菌体细胞完整结构。 The cell wall Whole Peptidoglycan (WPG) of Bifidobacterium bifidum Bif1101 was extracted and purified as follows: (1) Treating the bacteria cells with Triton X-100. (2) Separating the insoluble residue by high speed centrifugation. (3) Digesting the insoluble residue with trypsin, α-chymotrypsin, pepsin, Pronase E, DNase and RNase. (4) Delipoiding the digested residue by successive refluxing with methanol, methanol chloroform and chloroform. (5) Treatin residue with 0.01N H 2SO 4. The purified WPG was dialyzed and lyophilized. The results of chemical analysis indicated that WPG was consisted of polysaccharide and peptide, electeonmicroscopic observation of WPG showed that WPG had the physically intact cell wall structure of the whole cells. From these results above, isolating WPG from whole cell without being subjected to physically destructive methods provide a method for extract whole WPG.
