摘要
目的:建立大鼠血浆中麦冬多糖MDG-1的高效凝胶色谱测定方法。方法:血浆样品经30%三氯乙酸沉淀蛋白后,以0.1 M NaH_2PO_4-Na_2HPO_4缓冲液(pH 7.0)为流动相,色谱柱为Shodex Sugar ks-802凝胶柱,采用荧光检测器进行检测。结果:该法线性范围为0.25~50μg·ml^(-1)(r=0.998 9),低、中、高3个浓度的回收率为93.78%~104.6l%,日内、日间RSD低于6.74%。结论:该法准确、灵敏,操作简单,可用于血浆中MDG-1的含量测定。
Objective: To develop a method for determination of Polysaccharide MDG-1 in rat serum by HPGPC. Method: Serum protein was precipitated with 30% trichloroacetic acid , The analysis was conducted with a Shodex Sugar ks-802 column, and 0.1 M Natrium dilydrogen phosphoric-natrium hydrogen phosphoric buffer ( pH 7 ) was Served as mobile phase with fluorescence detector. Result: The linear range of Polysaccharide MDG-1 was 0.25 -50 μg·ml^-1( r = 0.998 9) and the average recoveries were 93.78% - 104.61% at three different concentrations, The RSD of intra- and inter - day was less than 6.74%. Conclusion: This method is accurate, sensitive and suitable for the pharmacokinetic study of Polysaccharide MDG-1.
