肌醇六磷酸对结肠癌HT-29细胞分化的影响 被引量:1

EFFECT OF INOSITOL HEXAPHOSPHATE ON CELL DIFFERENTIATION OF HT-29 HUMAN COLON CARCINOMA

摘要 目的研究肌醇六磷酸(IP6)对结肠癌细胞株HT-29细胞周期的影响,探讨IP6诱导HT-29细胞分化的作用及其机制。方法将HT-29细胞与不同浓度(0、1.8、3.3 mmol/L)IP6共同孵育24、48、72 h后,收集细胞,使用流式细胞仪检测细胞周期,并用透射电镜观察细胞结构的变化。结果除1.8 mmol/L IP6作用24 h对HT-29细胞周期分布没有明显的影响外,其他各组G0/G1期细胞明显增多,S期、G2/M期细胞则相应减少。同时,IP6作用后细胞结构发生改变,出现核固缩、细胞器丰富、微绒毛增多等分化趋势。结论IP6能够调节HT-29细胞周期,阻止细胞从G0/G1期进入S期,从而诱导细胞分化。 Objective To study the effect of inositol hexaphosphate (IP6) on HT-29 cell differentiation and explore its mechanisms. Methods HT-29 cells were cultured with different concentrations of IP6 (0, 1.8, 3.3 mmol/L) for 24 h, 48 h, and 72 h. Flow cytometry was used to determine the cell cycle. Transmission electron microscopy (TEM) was used to observe cell structural changes. Results Cell population in phase G0/G1 was increased, and that in phase S and phase G2/M decreased after culturing with IP6 at all concentrations and time intervals except the group which was cultured with 1.8 mmol / L IP6 for 24 h and showed no significant changes. Uhrastructural observation found more karyopyknosis, abundant cellular organells and increased mierovillus within cultured ceils. Conclusion IP6 could regulate the cell cycle of HT-29 cell, and prevent ceil progression from stage G1 to S, which facilitates cell differentiation.
