日本医蛭中一种新抗凝血蛋白质的仿生亲和纯化及鉴定 被引量:14

摘要 目的从日本医蛭中快速提取和纯化生物活性蛋白质。方法利用仿生亲和介质库进行配体筛选和1步纯化。结果从日本医蛭中纯化出了1种具有抗凝血活性的新型低丰度蛋白质,命名为NLP-1(New Leech Protein-1)。通过进一步分析,其相对分子质量为13800,蛋白回收率可达到69.2%,其在血浆中含量为0.02 mg/mL时即可产生明显的抗凝血现象。利用MALDI-TOF-TOF分析,在蛋白质数据库中未找到相似蛋白质或多肽。结论从日本医蛭中纯化出的NLP-1蛋白质是1种新型的抗凝血物质,具有很高的开发研究价值。 Purpose To introduce a rapid method for isolation and purification of an anticoagulant from Hirudo nipponia. Methods A specific bioactive protein has been purified by a biomimetic ligand, which was screening from a library of biomimetic ligands. Results A new low-abundant protein which has the bioactivity of anticoagulation has been one-step purified from Hirudo nipponia, named as NLP-1 (New Leech Protein-1). By further analysis, its mass was determined as 13 800, its recovery of the protein could reach to 69.2 %, and its anticoagulation activity was obviously detected as 0. 02 mg/mL. By MALDI-TOF-TOF, no similar protein or peptide was found in Protein Data Bank. Conclusion The protein NLP-1 which is one-step purigied from Hirudo nipponia belongs to a new class of anticoagulants.
