摘要
目的探讨大蒜素对人乳腺癌细胞株(MDA-MB-231)的抑制作用。方法采用四甲基偶氮唑蓝(MTT)法测定不同浓度(5、10、20、40、80mg/L)大蒜素对增殖MDA-MB-231的抑制率。通过药物半数抑制浓度(IC50)计算软件计算IC50。相差倒置显微镜下观察细胞形态的改变情况。结果10、20、40mg/L组大蒜素对MDA-MB-231的增殖有明显抑制作用(P<0.01)。24、48、72hIC50值分别为16.50、8.28、7.82mg/L。相差倒置显微镜下观察可见MDA-MB-231细胞分裂相减少,部分细胞漂浮,残存细胞形态不规则,状态差,反光差,出现细胞碎片。16.50、8.28、7.82mg/L的大蒜素作用24、48、72h后,琼脂糖凝胶电泳可见DNA凋亡梯形条带。结论大蒜素可使MDA-MB-231的增殖受到抑制,促进肿瘤细胞凋亡,具有抗肿瘤作用。
Objective To investigate the inhibiting effect of allicin on MDA-MB-231. Methods MDA-MB-231 cells in culture medium were treated with allicin at concentrations of 5,10,20,40 and 80 mg/L, respectively. The inhibitory rate of the cells was measured by MTT assay. The IC50 was drawed by the software of IC50. Morphology of cell was observed by the phase microscope. Results Allicin inhibited the growth of MDA-MB-231 significantly at 10,20,40 mg/L concentrations(P〈0.01). The 50% inhibiting concentration(IC50) of 24,48,72 h was 16.50,8.28, 7.82 mg/L, respectively. It could be both seen that the division of MDA-MB-231 cell was reduced, some ceils were floated, and seen irregular shape, poor state, poor reflecting and cell debris in the remnants by the phase microscope. There were DNA ladders on agarose gel electropho-resis after the cells were treated with allicin at concentrations of 16.50,8.28,7.82 mg/L for 24, 48 and 72 h,respectively. Conclusion Allicin can efectively inhibit the growth and induce apoptosis in MDA-MB-231 ,and it has anti-tumor effect.
