复方芍根口服液防治大鼠放射性食管炎氧化与抗氧化作用研究 被引量:5

The Study on the Items of Oxidation and Anti-oxidation in the Rats with Radiation Esophagitis Treated with Compound White Peony Root Oral Liquids

摘要 目的研究急性放射性食管炎大鼠经复方芍根口服液治疗后,大鼠食管羟自由基(OH.)、一氧化氮(NO)及血清一氧化氮(NO)水平和过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-PX)活性变化,分析其作用机制。方法将96只Wistar大鼠随机分为8组,分别为正常对照组(不做任何处理),单纯放射一组(放射后第7天处死),单纯放射二组(放射后第14天处死),复方芍根口服液正常剂量预防组(从放射开始给药),复方芍根口服液大剂量预防组(从放射开始给药),复方芍根口服液正常剂量治疗组(从放射后第7天给药),复方芍根口服液大剂量治疗组(从放射后第7天给药),西药治疗对照组(从放射后第7天给药)。在各实验结束点,留取大鼠血清和全长食管。检测大鼠食管组织匀浆液中OH.、NO含量,CAT、GSH-PX活性及血清NO水平。结果大鼠经43Gy^(60)Co-γ射线于食管局部单次照射后第14天,食管组织中OH和NO含量明显上升(P<0.05);CAT和GSH-PX活性水平也显著增高(P<0.01)。经复方芍根口服液治疗后,食管组织中的OH.和NO含量与单纯放射组相比明显下降(P<0.05),尤以复方芍根口服液大剂量治疗组下降明显(P<0.01);CAT和GSH-PX活性明显降低(P<0.05)。各组大鼠血清中NO水平差异无显著性(P>0.05)。西药治疗组大鼠与单纯放射组相比,食管组织中的NO含量及CAT和GSH-PX活性也明显下降(P<0.05),以NO含量下降显著(P<0.001)。结论复方芍根中药口服液有助于降低放射性食管炎大鼠食管食管组织中OH.和NO水平以及CAT和GSH-PX)活性,进而发挥保护食管的作用。 Objective To study the content of the hydroxyl radical ( OH ) and NO, the activity of CAT and GSH - PX in rats' esophagus and NO level in serum from the Wistar rats radiated for esophagitis area treated with compound white peony root oral liquids (cWPROL). Methods 96 Wistar rats were divided into eight groups. Group 1, the normal group, were without any radiation and treatment; Group 2, radiation group 1, killed on the seventh day after being radiated by 43Gy ^60Co-γ ray; Group 3, radiated group 2, killed on the fifteenth day after being radiated; Group 4 given cWPROL (normal dose) for prevention at the same time of radiation; Group 5 given cWPROL( high dose) for prevention at the same time of radiation; Group 6 were treated with cWPROL (normal dose) from the seventh day after radiation; Group 7 were treated with cW- PROL (high dose) from the seventh day after radiation; Group 8 were treated with Western medicine from the seventh day after radiation. On every experimental finished point, the blood serum and full - length esophagus of rats were kept for examination. The content of hydroxyl radical ( OH ) and NO, and the activity of CAT and GSH - PX in rats' esophagus tissues were determined. The content of NO in rats' blood serum was also analyzed. Results The content of hydroxyl radical ( OH ) and NO and in Wistar rats esophagus tissue increased significantly compared with that of the normal rat on the 14th day radiated by 43 Gy 60 Co -γ ( P 〈 0.05 ). The activity of CAT and GSH - PX increased significantly ( P 〈 0.01 ). However, the content of hydroxyl radical ( OH ) and NO, and the activity of CAT and GSH - PX in the esophagus tissue of rat treated with cWPROL decreased significantly compared with that of the rat in radiated group 2 ( P 〈 0.05 ), the rat treated with cWPROL of high dose from the seventh day after radiation ( P 〈 0.01 ). The content of NO in blood serum was no significantly different among all of the groups ( P 〉 0.05 ). The content of NO and the activity of CAT and GSH - PX in the esophagus tissue were reduced in the rats treated with Western medicine ( P 〈 0.05 ) compared with that of the rats in radiation group 2, especially the marked decrease in the content of NO (P 〈 0, 01 ). Conclusion cWPROL can reduce the content of hydroxyl radical ( OH ) and NO the activity of GSH- PX and CAT in esophagus tissue of rat radiated with ^60Co -γ ray, which maybe is the mechanism for the treatment of inflammatory esophagus induced by radiation.
