摘要
目的:探讨近年用来治疗恶性肿瘤的视黄酸与IL-1β、IL-4的相互作用对人肺癌细胞株Α549分泌补体C3及Β因子的影响。方法:分别用ELISA和RT-PCR检测培养上清中补体C3及B因子的水平及细胞补体C3及B因子mRNA转录。结果:ELISA检测结果表明IL-1β增强A549细胞分泌补体C3及B因子;IL-4增强A549细胞分泌C3,但对B因子没有影响,但其上调IL-1β诱发的Α549分泌C3,下调Α549分泌B因子;视黄酸对Α549细胞分泌补体C3及B因子没有影响,但它增强IL-1β和IL-4诱导的A549分泌补体C3及B因子分别是IL-1β和IL-4组的1.44、1.89,1.13、1.74倍。RT-PCR分析补体C3及B因子mR-NA的结果与蛋白水平相似。结论:IL-1β、IL-4和视黄酸相互作用影响人肺癌细胞株A549细胞分泌补体C3及B因子,调节补体系统,对杀伤肿瘤细胞有一定作用。
Objective :To explore the effect of retinoic acid (RA) interacting with IL- 1, IL- 4 on C3 and factor B (Bf) secreted by human lung cancer ceils line A549. Methods:The levels of C3 and Bf in culture supernatant and the expression of C3 and Bf mRNAs in cell were detected by ELISA and RT - PCR respectively. Resuits:ELISA results showed that IL - 1β enhanced the secretion of C3 and Bf of A549 ceils ; IL - 4 enhanced the secretion of C3 of A549 ceils and had no effect on Bf, but it could up - regulate the secretion of C3 of A549 ceils induced with IL - 1β and down - regulate the secretion factors of A549 ceils. Retinoic acid had no effect on secretion of C3 and Bf of A549 ceils, but enhanced the secretion of C3 and Bf of A549 ceils induced by IL - 1β and IL - 4. The levels of C3 and Bf secreted by A549 ceils induced with IL- 1β and IL- 4 were 1.44, 1.89, 1.β, 1.74 times of untreated group without RA. RT- PCR analysis showed that mRNA of C3 and Bf resembled with protein level. Conclusion:RA interac- ting with IL - 1β and IL -4 can effect the secretion of C3 and Bf of A549 ceils line of lung cancer, regulate the complement system and kill the tumor ceils.
