天花粉蛋白新有效组分的纯化及相对分子质量测定 被引量:5

Purification and determination of molecular weight of new active trichosanthin components

摘要 目的:研究从新鲜栝楼块根中分离纯化天花粉蛋白新有效组分的方法,测定新有效组分的相对分子质量,并对野生型及人工培植型植物来源的新有效组分进行对比研究。方法:采用粉碎、盐析、透析和离子交换层析、分子筛层析方法分别从野生型及人工培植型新鲜栝楼块根中分离纯化天花粉蛋白新有效组分,应用LDI-1700激光解吸电离飞行时间质谱仪测定其相对分子质量。结果:从野生型新鲜栝楼块根中纯化的天花粉蛋白新有效组分的相对分子质量为33388.9,产率为1.2mg.g-1;从人工培植型新鲜栝楼块根中纯化的天花粉蛋白新有效组分的相对分子质量为32116,产率为0.3mg.g-1。两者的相对分子质量高于已应用于临床的天花粉蛋白针剂(相对分子质量为26000)及从美国产的栝楼中分离纯化出的TAP29(相对分子质量为29000),野生型产率高于人工培植型的产率。结论:分别从野生型与人工培植型新鲜栝楼块根中纯化的天花粉蛋白新有效组分的相对分子质量及含量不同,且两者的相对分子质量亦不同于天花粉蛋白针剂及TAP29。
