摘要 目的探讨绒毛组织培养及染色体制备的理想方法。方法将73例标本用不同的消化酶处理后培养,应用原代传位法进行收获和染色体制备。结果经不同消化酶处理后,绒毛细胞培养时间具有显著相关性,但与核分裂相的数目无相关性。73例中,68例培养成功,成功率93.15%,检出异常核型30例。结论应用不同比例的胶原酶和胰酶的混合酶解液消化处理绒毛细胞后培养,用原代传位法进行染色体制备,具有效率高、组织细胞生长快、细胞生长周期短、收获中期分裂相不受影响和染色体形态保持良好等优势。 Objective To study the relationship of different kinds of digestive enzyme with villiform cell culture and chromosome preparation. Method Each specimen of 73 cases was treated with different kinds of digestive enzyme and villiform cell and harvested by the original cell division method. Results The time of villiform cell culture was obviously related to the kinds of digestive enzyme,but not to the number of karyotypes. The villiform cells of 68 specimens(93.15%) were cultured successfully and 30 abnormal karyotypes were tested. Conclusion The method of treating specimens with the mixture of different proportion of collagen enzyme and pancreas enzyme and preparing chromosome by the original cell division is of high efficiency, rapid growth of the cells and favorable karyotypes.
