高效液相色谱法测定南葶苈子饮片中芥子碱硫氰酸盐含量 被引量:8

摘要 目的采用高效液相色谱(HPLC)法测定南葶苈子饮片中芥子碱硫氰酸盐的含量。方法色谱柱为Phenomenex-C18柱(250mm×4.6mm,5μm),流动相为乙腈-0.08mol/LKH2PO4溶液(11∶89),检测波长为326nm,流速为1.0mL/min,柱温为室温。结果芥子碱硫氰酸盐进样量在0.944~4.720μg范围内与峰面积线性关系良好,加样回收率为98.85%,RSD=0.61%(n=6)。结论HPLC法操作简便、准确,具有很好的重现性和稳定性;炮制对南葶苈子饮片中芥子碱硫氰酸盐的含量有明显影响。 Objective To determine the sinapine thiocyanate in the Processed Pieces of Semen Lepidii by HPLC. Methods phenomenex- C18 column (250 mm x4.6 mm,5μm) was used,the mobile phase was acetonitrile -0.08 mol/L KH2PO4 (11 : 89). The flow rate was 1.0 mL/min and detection wavelength was at 326 nm. Results The linear range of sinapine thiocyanate was 0. 944-4. 720 μg, the recovery rate was 98.85%, RSD =0.61% (n =6). Conclusion This method is simple, rapid and accurate with good reproducibility. It is suitable for the quality control of the processed Pieces of Semen Lepidii. Processing has a significant influence on the concentration of sinapine thiocyanate in the processed pieces of Semen Lepidii.
