摘要
[目的]探讨溴乙锭(EB)与脱氧核糖核酸(DNA)的相互作用。[方法]在0.05mol/LNa2HPO4-0.05mol/LNaH2PO4-0.10mol/LNaCl缓冲溶液(pH=7.30)中,研究EB在核苷三磷酸修饰电极及单链DNA(ssDNA)和双链DNA(dsDNA)修饰电极上的电化学行为。[结果]在dsDNA和ssDNA均可使EB的氧化峰负移,核苷三磷酸对峰电位改变与dsDNA和ssDNA相似。[结论]EB与DNA的相互作用是EB接受碱基的电子形成∏-∏共轭造成的。
[ Objective ] To study the interaction between ethidium bromide ( EB ) and DNA. [ Methods ] The electrochemical behavior of EB at nucleoside triphosphate, single chain DNA ( ssDNA )and double chain DNA ( dsDNA )modified electrodes in 0.05mol/L Na2HPO4-0.05 mol/L NaH2PO4-0.10 mol/L NaCl buffer solution( pH=7.30 )was studied. [ Results ] The dsDNA and ssDNA could shift the oxidized potential of EB negatively, the shifted potential of EB at dATP, dCTP, dGTP and dTTP modified electrode was similar to that of dsDNA and ssDNA. [ Conclusion ] The interaction between EB and DNA was ascribed to the formation of П-Пconjugate between EB and bases of DNA.
