摘要
目的:探讨苦荬菜组织培养和植株再生条件。方法:以苦荬菜叶片为外植体,应用正交设计方法,对愈伤组织的诱导和分化培养基进行优化筛选。结果:最佳的愈伤组织诱导培养基为MS+2,4-D 1.5 mg.L-1+6-BA1.5 mg.L-1+NAA1.0 mg.L-1+IBA1.5 mg.L-1+KT1.5 mg.L-1,最佳的不定芽诱导培养基为MS+2,4-D 0.2 mg.L-1+6-BA0.5 mg.L-1+NAA 0.5 mg.L-1+IBA 0.5 mg.L-1+KT0.5 mg.L-1,不定芽经附加IBA0.1 mg.L-1的1/4 MS培养基生根后移栽成活,获得再生植株。结论:建立了一套有效的苦荬菜组织培养再生体系。
Objective: To study the callus induction and plant regeneration of Ixeridium sonchifolium. Method: By using the orthogonal experiment design, the medium for callus induction and plant regeneration was optimized. Result: The optimal medium for callus induction was MS+2,4-D 1.5mg·L^-1+6-BA 1.5mg·L^-1+NAA 1.0mg·L^-1+IBA 1.5mg·L^-1+KT 1.5mg·L^-1, the optimal medium for inducing adventitious bud was MS+2,4-D 0.2mg·L^-1+6-BA 0.5mg·L^-1+NAA 0.5mg·L^-1+IBA 0.5mg·L^-1+KT 0.5mg·L^-1. Plantlets were rooted on 1/4MS medium supplemented with different concentrations of IBA, and high rooting and survival was achieved when the IBA concentration was 0.1 mg·L^-1. Conclusion: An efficient system for plant regeneration of I. sonchifolium was established.
