摘要
目的研究雌黄纳米粒对慢性粒细胞白血病急变期K562细胞端粒酶活性的影响,初步探讨雌黄纳米粒抗肿瘤作用的分子机制。方法采用端粒重复序列扩增-聚丙烯酰胺凝胶电泳银染法检测雌黄纳米粒对K562细胞端粒酶活性的影响,同时与传统剂型的雌黄作用进行比较。结果雌黄纳米粒对K562细胞的端粒酶活性有强烈的抑制作用,与未加药组及传统剂型的雌黄组相比,雌黄纳米粒组端粒酶活性下降更明显。结论雌黄纳米粒能显著下调K562细胞端粒酶活性,效果明显优于传统剂型的雌黄,可能是其发挥抗肿瘤作用的重要机制之一。
Objective To study the change in telomerase activity during proliferation inhibition of leukemia K562 cells induced by orpiment nanoparticles and further explore the molecular mechanism of the antitumor effect of orpiment nanoparticles. Methods Methyl thiazolyl tetrazolium (MTT) assay examines the growth inhibition induced by orpiment nanoparticles in vitro. TRAP -PAGE silver stain technique was performed to detect the telomerase activity of K562 cells treated with orpiment nanoparticles for 48 hours, compared with that treated with traditional orpiment at the same time. Results Orpiment nanoparticles significantly inhibited the telomerase activity of K562 cells, with a much better effect than that of traditional orpiment. Conclusions Orpiment nanoparticles can inhibit the telomerase activity of K562 cells, which may be one of important mechanisms in the anticancer effect of orpiment nanoparticles.
