摘要
以离体的芝麻花粉为实验材料,研究了不同的培养基组分以及超声波处理对芝麻花粉萌发的影响。实验结果表明:蔗糖对于芝麻花粉的萌发有重要作用,当蔗糖浓度为35%时,芝麻花粉内外的渗透压达到平衡,有利于花粉萌发;培养基内添加适量的硼酸和氯化钙能有效地促进花粉萌发,但超过一定浓度时起抑制作用;培养基以35%的蔗糖,40 mg/L的硼酸,200 mg/L的氯化钙较为适宜,花粉的萌发率可以达到58%左右;超声波处理的功率、处理时间以及处理次数对芝麻花粉的萌发率和破碎率有显著影响。
Effects of medium composition and ultrasonication treatment on the germination of sesame pollen were studied through the in vitro pollen cultivation. The results showed that sucrose, boric acid and calcium chloride were very important for pollen germination. Pollen germination reached 58% in the medium containing 35% of sucrose, 40 mg/L H3BO3 and 200 mg/L CaCl2, respectively. Orthogonal design was employed to determine the parameters ofultrasonication treatment. It was also revealed that treatment power, time and number ofultrasonication had significant effects on the broken rate and germination rate of sesame pollen.
