摘要
目的:探讨随机扩增多态性DNA(RAPD)检测技术的特点。方法:分析影响RAPD标记的各种因素,总结该技术的优点和不足。结果:RAPD标记方法已经广泛用于遗传育种、基因诊断、生物系统学的研究,并显出独特的优点。DNA模板质量与浓度、TaqDNA聚合酶、引物设计及浓度、缓冲体系、MgCl2、dNTP浓度、PCR反应体积等均为该技术的影响因素。结论:对于每一种物种都应先进行体系优化,确保RAPD实验结果稳定准确。
Objective: To expore the characteristics of the detect technique of random amplified polymorphic DNAs (RAPD). Methods: Analyzed the factors affecting RAPD technique and summarized merits and deficients of that technique. Results: RAPD technique is widely used in studies of heredity breeding, gene diagnosis and biosystematics, showing some unique merits. Concentration and purity of the genomic DNA, design and cocentration of primers, concentration of Taq DNA polymerase, buffer system, MgCl2 and dNTP, as well as PCR reaction volume are all the affecting factors. Conslusion: Every species should be optimized for RAPD system and ensure the stable and accurate experimental result.
