摘要
目的评价中药复方—连归清心耐缺氧及抗心肌缺血损伤的效果,为临床应用提供药理学依据。方法:采用密闭缺氧实验,观察连归清心对小鼠耐缺氧存活时间的影响;采用冠脉结扎致急性心肌缺血模型,观察连归清心对大鼠心电图ST段移位、心肌梗死范围及血清心肌酶活力的影响。结果中、高剂量连归清心组小鼠密闭缺氧的存活时间分别为(974.0±59.5)s和(1004.8±125.5)s,明显长于对照组(861.8±114.5)s(P<0.05),心肌梗死面积比对照组分别减小36.8%与58.1%(P<0.01);连归清心高、中、低剂量可显著降低冠脉结扎后不同时间段心电图ST段的抬高程度(P<0.05),并减弱血清AST、LDH-L与CK-MB的活力(P<0.05)。结论连归清心可增强小鼠耐缺氧能力,并对缺血心肌具有明显的保护作用。
Objective To study the effects of Chinese medicine—Lianguiqingxin(LGQX) on hypoxia tolerance and acute myocardial ischemia,so as to provide pharmacological basis for further clinical study.Methods Hypoxia tolerance test was performed by placing the mice into a closed bottle to measure their survival time.Myocardial infarction model was made by ligation of left anterior descending coronary artery of rats,and myocardial infarction area(MIA),changes of ECG and segment ST,as well as myocardial enzyme activity were observed.Results Both 2.40 and 4.80 g·kg^-1·d^-1 of LGQX could prolong survival time of mice under closed hypoxia condition((974.0±59.5)s,(1004.8±125.5)s respectively vs(861.8±114.5)s,P〈0.05).The MIA in 2.40 g·kg^-1,and 4.80 g·kg^-1 group were reduced by 36.8%(P〈0.01),58.1%(P〈0.01) respectively.The elevation of ST segment in ECG at different time points,and the decrease in activities of serum AST,LDH,CK-MB were also observed(P〈0.05) in acute myocardial infarction rats treated with LGQX(1.20~4.80g·kg^-1·d^-1). Conclusion LGQX can enhance anti-hypoxic effect of mice,and has obvious protective effect on ischemic myocardium.
