摘要
目的探讨单独及联合应用川芎嗪与针刺对庆大霉素致耳聋豚鼠螺旋神经节细胞凋亡活性及相关蛋白Bax和Bcl-2表达的影响。方法腹腔注射庆大霉素100mg·kg^(-1)·d^(-1),连续10天,建立感音神经性耳聋豚鼠模型。然后动物随机分组,分别单独或联合予以川芎嗪与针刺治疗。疗程结束后处死动物,应用普通病理组织学技术确定各组耳蜗病理变化,TUNEL标记和免疫组化法观察比较不同疗法干预治疗对实验动物螺旋神经节内神经细胞凋亡活性及凋亡相关蛋白Bax和Bcl-2表达的影响。结果针刺联合川芎嗪组动物的耳蜗病理形态学明显改善,凋亡细胞数量明显减少,Bax/Bcl-2的表达比例降低显著。结论单独及联合应用针刺与川芎嗪均可明显拮抗庆大霉素致耳聋豚鼠螺旋神经节内神经细胞的凋亡活性及相关蛋白Bax和Bcl-2的表达,尤以针刺"听宫"、"外关"及"翳风"等穴的联合效应明显。
Objective To investigate the protective effects of Ligustrazine and acupuncture used separately or together on apoptotic activity and expression of apoptosis-related genes bcl-2 and bax in spiral ganglion cells of guinea pigs with gentamicin-induced hearing loss. Methods The model of sensorineurel hearing loss was prepared by consecutive intra-peritoneal injection with gentamycin（ 1 00mg/ kg·d） for 10 days among guinea pig. Then, the animal models were randomly divided into several groups receiving different treating procedures with Ligustrazine and acupuncture used separately or together respectively. Followed was determination on the effects of these intervening procedures on the apoptotic activity and expressing levels of apoptosis-related proteins Bcl-2 and Bax in spiral ganglion cells of theses experimental animals by TdT-mediated DUTP-biotin nick end labeling（TUNEL）and immunohistochemical techniques respectively by the end of therapeutic course with the animals put into death and all tissue samples collected. Results When compared with that of other groups of guinea pigs treated by different intervening procedures, significant improvement in the pathohistology of inner ear was found in the group of animals received the combined therapy with Ligustrazine and acupuncture, with markedly decreased number of apoptotic spiral ganglion cells and reduced expressing ratio of Bax/Bcl-2 in these spiral ganglion cells. Conclusions Ligustrazine and acupuncture can effectively inhibit the apoptotic activity and regulate the expressing levels of apoptosis-related proteins Bcl-2 and Bax in spiral ganglion cells of guinea pigs with gentamicin-induced hearing loss, no matter how they are used,either used alone or in a combined way,especially when the drug given in a combined way with acupuncturing to the points of Tinggong,Waiguan, Yifeng and so on.
