摘要
以沈农糯玉米为试材,改良MS培养基为基本培养基,研究了不同浓度的2,4-D对玉米幼胚愈伤组织的诱导效果,筛选出最适的2,4-D浓度为4 mg/L。4 mg/L 2,4-D配合使用较低浓度的6-BA(0.5 mg/L)不仅可以提高愈伤组织的诱导率,而且胚性愈伤组织的诱导率也有所提高。培养基中同时加入700 mg/L水解酪蛋白和1 000 mg/L的L-脯氨酸可显著提高胚性愈伤组织的诱导率;甘露醇和AgNO3对胚性愈伤组织诱导率提高无明显作用,但对改善胚性愈伤组织的质量起重要作用。
Study on the effects of phytohormones and appendages to inducing callus production of maize immature embryo was conducted so as to select the fittest concentration by the material of Shennong nuo and with the help of improved MS medium. As a result, the best 2,4-D concentration of 4 mg/L is perfectly supplemented by the lower 6-BA concentration of 0.5 mg/L, which should inhance not only inducing rate of callus but also embryogenic callus. At the same time, it is discovered that adding 700 mg/L caseine hydrolysate and 1 000 mg/L L-Proline into above mediam should significantly improve inducing rate of embryogenic callus, but D-Mannitol and AgNO3 have no significant effects on inducing rate of embryogenic callus except for improving quality of callus.
