双向凝胶电泳和质谱技术在视网膜蛋白质表达研究中的应用

摘要 目前对视网膜功能的研究大多停留在形态学和基因组水平,这些研究并不能反映基因转录后的调控、蛋白质表达水平的变化以及蛋白质的翻译后修饰等信息,且细胞内mRNA水平和蛋白质的丰度并不完全一致,所以从蛋白质水平上研究视网膜的功能显得尤为重要。与DNA的提取和聚合酶链反应(polymerase chain reaction,PCR)不同,组织或器官蛋白质用于蛋白质组学研究时,其蛋白质的抽提方法、双向电泳和质谱鉴定的条件都不完全相同。
