摘要
目的:探讨左旋精氨酸与急性胰腺炎(AP)大鼠胰腺腺泡细胞凋亡的关系.方法:以蛙皮素制造AP模型,观察不同剂量L-Arg治疗AP大鼠后,胰腺组织中一氧化氮合酶(NOS)活性、一氧化氮(NO)的含量、胰腺腺泡细胞凋亡及胰腺组织病理改变.结果:AP胰腺组织中NOS活性较正常胰腺组织中升高,而此时胰腺组织NO浓度反而降低,低剂量L-Arg(75,150mg/kg)使胰腺组织中NO浓度升高致接近正常组水平,促进了胰腺腺泡细胞凋亡,减轻胰腺病理损害;高L-Arg剂量(1200,2400mg/kg),胰腺组织中NO浓度升高超出正常组,胰腺腺泡细胞凋亡减少,胰腺病理损害加重.结论:L-Arg对AP的的双重作用与NO的生物活性有关.小剂量可通过诱导胰腺腺泡细胞凋亡,而起保护作用,大剂量则有害.
AIM: To investigate the effects of different doses of L-arginine on pancreatic acinar cell apoptosis in rats with acute pancreatitis (AP). METHODS: The model of AP was estab- lished in rats by 50 μg/kg caerulein for 2 h, and the rats in 6 treatment groups received injection of different doses of L-arginine (75, 150, 300, 600, 1200, 2400 mg/kg ) at the beginning of the 4th hour. The alterations of pathology, NOS activity, NO2^ - / NO3^- concentration, and apoptosis index were evaluated at the end of 12th hour. RESULTS: The activity of NOS increased in the pancreas of rats with AP, while NO2^-/NO3^- concentration decreased. Lower doses of L-arginine (75, 150 mg/kg) promoted the acinar cell apoptosis and decreased the pathological scores, while higher doses of L-arginine ( 1200, 2400 mg/kg) reduced acinar cell apoptosis and increased the pathological scores. CONCLUSION: The lower dose of L-arginine has a protective effect against AP, while higher dose of L-arginine may be harmful.
