摘要
目的:培养不同脑部和脑区的微血管内皮细胞.方法;取健康兔或猪的不同脑部或脑区组织,先经胰酶初步消化、匀浆、尼龙筛网过滤和右旋糖酐分离后,用胶原酶消化获取的微血管并进行培养.用Ⅷ因子相关抗原抗血清进行免疫组织化学染色鉴定.结果:培养的细胞呈单层贴壁生长,Ⅷ因子相关抗原染色阳性.原代培养的内皮细胞至少存在两种表型,其中一种可传代培养.结论:建立了一种简便培养脑徽血管内皮细胞的方法.细胞表型的不同,可能与其功能异质性有关.
Objective:To culture regional brain microvascular endothelial cells. Methods: Brain stem, cerebellum, regional cerebral cortex tissue from healthy rabbits or porcines were digested by trypsin first, and then filtered by nylon mesh, separated by dextran, respectively. The microvasculature obtained from above procedures were digested again by collagenase and cultured. Results: The cultured endothelial cells displayed monolayer growth and were stained positively for factor Ⅷ relative antigen. There were two phenotypes in primary cultured microvascular endothelial cells at least, and one of them could be continuously cultured. Conclusion: It is a simple method to culture endothelial cells from brain microvasculature and the difference of phenotypes may be related to their function.
