厚皮甜瓜高效再生体系的建立 被引量:17

摘要 以优质厚皮甜瓜品种鲁厚甜2号和鲁厚甜4号为材料,系统研究了以子叶为外植体的厚皮甜瓜组培高效再生体系。结果表明:两个甜瓜品种在不定芽和愈伤组织诱导上存在着较大的差异,在MS+6-BA 1.0 mg/L+IBA 0.5 mg/L的分化培养基上,鲁厚甜2号不定芽诱导率最高达92.6%;而鲁厚甜4号在MS+6-BA 2.0 mg/L+IBA 0.2 mg/L中达94.3%;诱导出的不定芽丛的伸长以MS+KT 0.2 mg/L为最佳;两个甜瓜品种在1/2MS+IBA 0.4 mg/L中均获得良好的生根效果。另外,保持高温、高湿是甜瓜再生苗移栽成活的重要条件。 An efficient regeneration system of thick-skinned muskmelon had been set up using the cultivars Luhoutian2 and Luhoutian4 cultured in the mediums with different hormone combination. The results showed that MS+6-BA1.0mg/L+IBA0.5mg/L was the most suitable medium for Luhoutian2 to induce adventitious buds while MS+ 6-BA 2. 0rag/L+IBA0.2mg/L for Luhoutian4; and their most inductivity were 92.6 % and 94.3 %, respectively. The optimization medium for adventitious buds to lengthen was MS+KT0. 2mg/L. Both Luhoutian2 and Luhoutian4 had good rooting effect on the medium 1/2MS IBA0.4mg/L. In addition it was important for transplanting survival of regeneration seedlings to keep high temperature and high humidity at the same time.
