摘要
目的探讨丹皮酚(Paeonol,Pae)对人脑胶质瘤细胞株U251增殖和凋亡的影响。方法采用MTT法检测丹皮酚对体外培养的人脑胶质瘤细胞株U251的增殖抑制作用,用透射电镜和流式细胞仪观察Pae对U251细胞的诱导凋亡作用。结果Pae在31.25—250mg/L浓度范围内对U251细胞的增殖均有抑制作用,呈现明显的量效和时效依赖关系。诱导电镜下可见肿瘤细胞发生凋亡改变。Pae在31.25—250mg/L浓度下均可诱导U251细胞发生凋亡,其浓度越高、作用时间越长,凋亡率越高,有明显的时效和量效关系。结论Pae对有抑制人脑胶质瘤细胞株U251的增殖和诱导其发生凋亡的作用。
Objective To investigate the effect of paeonol on proliferation and apoptosis of U251 in vitro. Methods The inhibitory effect of paeonol on the proliferation of U251 cells was assayed by MTT in vitro, the morphology of apoptotic cells was observed by transmission electron microscope, and flow cytometry was used to analyze the apoptosis of U251 cells induced by paeonol. Results The proliferation of U251 cells was inhibited by paeonol with the concentration of 31.25 mg/L to 250 mg/L in dose and time dependent manner Typical apoptotic changes in U251 cells were observed by transmission electron microscope. By FCM methods, the apoptotic rates of U251 cells were increased when the concentrations of paeonol increased from 31.25 mg/L to 250 mg/L, which also showed obvious time effect relationship. The distribution of cell cycle was also changed after treated by paeonol. The number of cells in S phase increased and that in G0/G1 phase and G2/M phase decreased and the cell cycle was incased in S phase. Conclusion The proliferation of U251 cell can be inhibited and its apoptosis can be induced by paeonol.
