摘要
目的:观察血小板经不同mPEG修饰后功能变化情况,并分析其原因。方法:分别用mPEG-BTC、mPEG-SPA对血小板进行化学修饰;流式法检测修饰前后血小板CD62P荧光强度变化;并对血小板的聚集、黏附及释放功能进行检测。结果:经mPEG-BTC、mPEG-SPA修饰后,血小板CD62P表达率由修饰前的(12.77±2.87)%分别降至0和(4.12±1.28)%;聚集能力由修饰前的(1.43±0.74)%分别升至(3.86±1.21)%和(3.84±1.12)%;黏附能力由修饰前的(30.2±5.59)%分别升至(45.5±2.72)%和(45.2±3.99)%;而血小板Ca2+释放能力修饰前后差异无统计学意义。结论:使用mPEG-SPA对血小板进行化学修饰可影响血小板的功能,是一种较为理想的修饰剂。
Objective:To investigate the metergasis of platelets camouflaged by methoxy polyethylene glycols(mPEG) with different active groups, and to analyze its mechanism, nethods:Platelets were camouflaged by mPEG benzotriazole carbonate(mPEG-BTC) or succinimide of mPEG propionic acid(mPEG-SPA). The activated molecule CD62P of platelets was detected by flow cytometry and the platelet functions of aggregation, adhesiveness, and release were detected as well. Results:After the platelets were camouflaged by mPEG-BTC or mPEG-SPA, the expression rate of CD62P dropped from pre-modificatoin ( 12. 77 ± 2. 87) % to post-modification 0 and (4.12 ± 1.28 ) %, respectively. But the ability of aggrega- tion was enhanced from pre-modificatoin ( 1.43 ± 0. 74) % to post-modification ( 3.86 ±1.21 ) % and ( 3.84 ±1.12 ) %, and the ability of adhesion was enhanced from pre-modificatoin (30.2 ±5.59 )% to post-modification (45.5 ± 2.72 )% and (45.2 ±3.99 ) % respectively. The ability of Ca^2+ release was not influenced by the modification. Conclusion: The effect on the metergasis of platelets camouflaged by mPEG-SPA was less than that by mPEG-BTC, mPEG-SPA was a suitable modifier to camouflage platelets.
