淡紫拟青霉几丁质酶的形成条件 被引量:5

摘要 采用摇瓶试验,研究了淡紫拟青霉几丁质酶的形成条件。在马铃薯煎汁、玉米粉、黄豆粉3种天然基质中,以马铃薯煎汁的产酶效果最好。蔗糖、果糖、山梨糖和可溶性淀粉等碳源的添加能增加酶的产量。在氮源方面,酵母膏、蛋白胨、尿素、精氨酸等能促进酶的形成。几丁质酶可为外源几丁质所诱导,当培养液中含有0.2%的粉状几丁质时,酶的产量显著上升。淡紫拟青霉形成几丁质酶的适宜培养基初始pH为6.0~6.5,培养时间以6~8d为佳。 The conditions that Paecilomyces lilacinus produced chitinase were studied by shaking culture. Among the three natural substances investigated, potato infusion was the most suitable for chitinase formation. When soluble starch, sucrose, fructose or sorbose were added into potato infusion, P. lilacinus increased the amount of both mycelia and chitinase. Chitinase was stimulated by nitrogen sources such as peptone, yeast extract or urea and inhibited by potassium nitrate, cysteine or ammonium sulphate. Chitinase was indued by chitin. When 2 g chitin powder was added into medium, the yield of chitinase grew significantly. Product of the enzyme was optimal at initial pH 6.0~6.5 and 6~8 days age of the mycelia.
