银杏叶提取物金纳多心停跳液对人心肌细胞bcl-2和bcl-xL基因表达的影响 被引量:3

摘要 目的探讨银杏叶提取物金纳多(简称金纳多)对体外循环人心肌细胞抗凋亡基因bcl-2和bcl- xL表达的影响。方法择期先天性心脏病患儿30例,随机均分为两组,治疗组在灌注St Thomas'停搏液(4℃)中加入金纳多(0.5 mg/kg),心脏完全停跳后开始心内手术;对照组单纯灌注St Thomas'停搏液(4℃)。在手术结束开放升主动脉前取心肌组织,采用RT-PCR技术在凝胶电泳成像基础上半定量检测bcl- 2和bcl-xL基因的表达水平。结果治疗组bcl-2和bcl-xL基因的表达显著高于对照组(P<0.05)。结论银杏叶提取物金纳多可促进人心肌细胞抗凋亡基因bcl-2和bcl-xL的表达。 Objective To investigate mRNA expression in the myocardium of Methods Thirty congenital heart disease the effects of Ginkgo biloba extract (Ginaton) on bcl-2 and bcl-xL patients underwent hypothermic cardiopulmonary bypass (CPB). patients were randomly assigned to 2 groups, the control group and the treated group. Patients in both groups received St. Thomas' cardioplegic solusion via radix aortae, while Ginaton (0.5mg/kg) was added in the treated group. Cardiac surgery was started after complete heart arrest. Myocardium was taken before the aorta ascendens was unblocked and mRNA expression of bcl-2 and bcl-xL in the ventricular tissue was detected by RT-PCR. Results The gene expressions of bcl-2 and bcl-xL were significantly higher in the treated group than those in the control group (P 〈 0.05). Conclusion Ginaton could promote the mRNA expressions of the antiapoptotic gene bcl-2 and bcl-xL in myocardium of patients underwent CPB.
