摘要
在pH=2.0的Britton—Robinson(B—R)缓冲液中,牛血清白蛋白(BSA)与四溴荧光素(1BF)形成复合物,在波长540nin处有最大吸收,比四澳荧光素本身的最大吸收红移25nm。对不同pH值及离子强度对其结合作用的影响进行了探讨,并研究了各种实验条件对反应体系的影响,确定了最佳反应条件,制订出BSA测定标准曲线。在四溴荧光素浓度为1×10^-5mol·L^-1时,其线性范围为0.05~12mg·L^-1,检出限为23μg·L^-1(S/N=3),方法稳定性好,检测灵敏度高。该方法用于实际样品的测定,结果令人满意。
The interaction of protein with Tetrabromo(R) fluorescein in B - R buffer was studied by spectrophotometry, the experiment conditions can affect their interaction. Based on this phentmmnon, a determination method of protein was established. Calibration curves under optimal conditions were prepared and a linear relationship of proteins exists in the concentration range of 0.05 ~ 12 mg · L^- 1 with the detection limit 23μg·L^ -1 ( S/N = 3). This method has been successfully applied for the determination of human serum albuinin.
