2ME2对全脑缺血大鼠HIF-1α的影响 被引量:5

Effect of 2ME2 on hypoxia-inducible factor 1α following the global cerebral ischemia in rats

摘要 目的探讨2-甲氧基雌二醇(2M E 2)对全脑缺血大鼠模型海马缺氧诱导因子-1α(H IF-1α)的影响。方法成年雄性SD大鼠120只,随机分为假手术组、缺血组和2M E 2组,后两组又分别分为6 h、12 h、24 h、48 h、72 h、5 d和7 d亚组。采用改良的Pu lsineli法制作全脑缺血大鼠模型。应用N issl染色检测大鼠海马神经元数,免疫组化方法检测H IF-1α蛋白表达。结果与缺血组比较,2M E 2组海马CA 1区神经元计数增加,H IF-1α蛋白表达降低,48 h^7 d亚组与缺血组相应时间点比较有统计学差异。结论严重全脑缺血损伤急性期应用2M E 2可抑制H IF-1α的过度表达,具有神经保护作用。 [Objective] To observe the effect of 2-methoxyestradiol (2ME2) on the hypoxia-inducible factor 1α (HIF-1α) following the global cerebral ischemia in rats. [Methods] 120 adult male Sprague-Dawley rats were randomly divided into 3 groups: sham group, global cerebral ischemia (GI) group and GI+ 2ME2 group, and the rats of GI group and GI+2ME2 group were sorted at 6 h, 12 h, 24 h, 48 h, 96 h, 5 d and 7 d after reperfusion. The 4 vessel occlusion (4-VO) global ischemia rat model was produced. Nissl staining was used to perform quantitative analysis of cell numbers. Immunohistochemistry staining provided the information of the distribution of positive cells to HIF-1α protein. [Results] The number of neuron in 48 h to 7 d groups from GI+ 2ME2 group was significantly higher than those from GI group. The level of HIF-1α in 48 h to 7 d groups from GI+2ME2 group was significantly lower than those from Gi group. [Conclusion] 2ME2 can restrain the elevation of HIF- 1α,which has brain protective effect on global cerebral, ischemia in rats.
