摘要
利用环糊精衍生物2-O-丙酮基-2-O-羟丙基-环糊精(2-AHP--βCD)作为手性选择剂,提出了一种毛细管电泳快速分离、测定柴胡中柴胡皂甙a,d对映体的方法.考察了手性选择剂浓度、缓冲液浓度及其pH值对分离效果的影响,确定了最佳分离条件.结果表明,在214 nm检测波长处,在硼砂缓冲溶液中,2-AHP--βCD能使柴胡皂甙a,d异构体在7 min内得到分离.利用该法测定了柴胡中柴胡皂甙a,d的含量,并进一步研究了方法的线性范围、检测限及精密度.
A fast capillary zone eleetrophoresis (CZE) method was developed for the separation and determination of saikosaponin a and d isomer in the Chinese herbal extract in Bupleurum. A highly water-soluble cyclodextrin (CD) derivative 2-O-Acetonyl-2-O-hydroxypropyl-β-CD (2-AI-IP-β-CD) was tested as an effective chiral selector for the capillary zone eleetrophoretic resolution of saikosaponin a and disomer. Detection at 214 nm with a system containing sodium borate buffer and 2-AHP-β-CD is found to be the most suitable approach for this analysis. The chiral resolution is strongly influenced by the concentration of the 2-AHP-β-CD, the con- centration of buffer and buffer pH. The compounds studied can be enantiomerically resolved in less than 7 minutes with no capillary treatment. Furthermore, the linearity range, quantitation limit for the crude drug extract in Bupleurum and the precision of the measurements were determined.
